چکیده

Small GTP-binding proteins act as switches that control diverse cellular processes. Those in the ADP-ribosylation factor (ARF) class direct key steps in intracellular protein traffic, whereas those in the Rho family regulate gene transcription as well as changes in cell motility and morphology [1,2]. Several groups have now produced evidence that these regulatory activities of Rho and ARF proteins may be mediated by phosphatidylcholine (PC)-specific phospholipase D (PLD). PLD generates phosphatidic acid (PA), which may be an intracellular mediator in its own right and which can be metabolized to form lipids with proven roles as both intercellular and intracellular messengers [3].

Intracellular signalling: Is PIP2 a messenger too?

The phospholipid phosphatidylinositol (4,5) bisphosphate (PIP(2)) has recently been shown to act downstream of the small G proteins Rac and Arf. Different effectors may be employed in each case, suggesting that PIP(2) has multiple signalling roles.

Small GTP-binding protein-coupled receptors.

Heterotrimeric GPCRs (G-protein-coupled receptors) form the largest group of integral membrane receptor proteins and mediate diverse physiological processes. In addition to signalling via heterotrimeric G-proteins, GPCRs can also signal by interacting with various small G-proteins to regulate downstream effector pathways. The small G-protein superfamily is structurally classified into at least ...

ARAP3 is transiently tyrosine phosphorylated in cells attaching to fibronectin and inhibits cell spreading in a RhoGAP-dependent manner.

ARAP3 is a GTPase activating protein (GAP) for Rho and Arf GTPases that is implicated in phosphoinositide 3-kinase (PI 3-kinase) signalling pathways controlling lamellipodia formation and actin stress fibre assembly. We have identified ARAP3 as a phosphorylated target of protein tyrosine kinases. In cells, ARAP3 was tyrosine phosphorylated when co-expressed with Src-family kinases (SFKs), upon ...

GTPase Signaling: Bridging the GAP between ARF and Rho

Membrane traffic and actin cytoskeleton dynamics are intimately linked, and GTPases of the Rho and ARF families may work together to regulate both. Recent studies have identified a family of GTPase activating proteins (GAPs) that contain both ARF-GAP and Rho-GAP domains, providing the first direct link between these two signaling pathways.

Cytohesin binder and regulator (Cybr) augments T cell receptor-induced NFAT-AP-1 activation through regulation of the JNK pathway

Cytohesin binder and regulator (Cybr also known as CYTIP, CASP, and PSCDBP) is a cytokine-induced gene preferentially expressed in haematopoietic tissues and in T helper (Th) 1 cells. Cybr protein associates with members of the cytohesin family, which are known ADP ribosylation factors GDP/GTP exchange factor (Arf)-(GEFs) and its functions appear to regulate lymphocyte adhesion and cell-cell co...
